Only once, has a party not nominated a sitting president, in 1852, and that was democrat Franklin Pierce who failed to receive his party's nomination.
So it's highly unlikely to happen now. But who knows?

Yes, in order to represent the D or R party one must receive their party's nomination. And there's no legal obligation to nominate a sitting president.
That said, one has to consider how much of a campaign advantage a sitting POTUS has before going with someone else. The optics of AF1 or their motorcade are things campaign contributions can't buy.

Biden has won his party's primary elections. Those delegates are pledged. There is no mechanism for anyone else unless "they" want to smother him with a pillow. Apparently, it's open season on your political opponents now because...democracy.
